Things you should know prior to setup:

Are you going to share any database tables accross sites?

Are end users going to be able to create their own site or admin only?

Are users to be shared accross sites?

Should users remain logged in when entering different domains (or subdomains, subdirectories)?

Once those questions are answered then you have a target to shoot at..

Get it set up without plugins first, then add as needed.

Don't expect the plugins to handle the fundamental setup

WPMU-DEV puts out great pllugins, and detailed support for such plugins is best found on their site.

Regardless of whether or not plugins are used to aid with multisite setup, it is important to note:

Although plugins can be very helpful in multi-site setup, the core functionality comes from Wordpress. Most results can be acheived through Wordpress if one knows which edits to make. My multisite setups are fully mapped and have all functionality without a plugin to aid in setup.

Sometimes plugins can actually prevent one from gaining knowledge necessary to continually increase in ability.
